Lee and Costlow (Methods in Enzymology 152, 633) describe the molecular titration assay for measuring the prevalence of specific transcripts. The method involves driving labeled RNA probes into RNA-RNA hybrids as assayed by nuclease protection. A constant amount of probe is titrated with increasing quantities of message. After nuclease digestion, hybrids are collected on a filter and counted. A plot of counts vs RNA concentration allows one to determine accurately the transcript prevalence. The authors claim the method to be the most sensitive RNA quantitation procedure. If anyone has had experience with the procedure, I would appreciate hearing from you. -- dn/dx Dept Molecular Biology diaz@aecom.yu.edu Dan Diaz Albert Grubelstein College of Osteopathy
