The key that I think your talking about is the one that you let the dog-thing out in the upper ishran castle. Mushroom land is found by tempting the large guy the the sword (to the left of the old man) to follow you back across the bridge. He will break it and leave a hole for you to fall through. This leads to mushroom land. Here you ask the snail about Karamoon to get back. You also obtain the bugle here which is used to cross the sea. Steve Harter.
