The six significant ORF's of Tn917 all occur on the same strand
and
are read from left to right. The start site of ORF1 is preceded by
a consensus promoter sequence (positions 420-425; 447-452) and a
Shine-Dalgarno sequence (positions 497-500). ORFs 4 and 6 are
also
preceded by potential promoter sequences.
ORFs 1-3 show strong sequence homology to the previously sequenced
erythromycin resistance determinant on S.sanguis plasmid pAM77.
ORFs 1-3 are flanked by a left terminal repeat, positions 94-166,
and an almost identical internal direct repeat, positions
1550-1622. The 3' terminus of Tn917 contains an inverted repeat,
positions 5313-5350, corresponding to the first 38 positions of
the
left terminus and internal repeat sequences.
A 20-bp region with 80% homology to the Tn3 resolvase is present
within the two directly repeating sequences. ORF4 also shows some
homology to Tn3 resolvase as well as some other recombination
enzymes.
There is no Shine-Dalgarno site preceding ORF5, so the translation
of ORFs 4 and 5 may be closely coupled. Also mutations within ORFs
5 and 6 indicate that these regions are essential for
transposition.
