The fusion (F) mRNA contains 1845 nucleotides, exclusive of
poly(A), has an unusually long (193-nucleotide) 5'nontranslated
region, and encodes an unprocessed Fo protein of 539 amino acids.
To establish the authenticity of the cDNA sequence, the F gene was
sequenced directly, in its entirety by dideoxynucleotide sequencing
of vRNA. This confirmed the cDNA sequence in general, but also
identified differences between the cDNAs and vRNA at eight
nucleotide positions (position 494, 614, 652, 1093, 1564, 1574,
1588, 1783). These are thought to represent variability introduced
during PF3 RNA synthesis (reported in [1]).

The complete nucleotide sequence of the phosphoprotein (P) mRNA of
human parainfluenza virus 3 (PIV-3) was derived from two cDNA
clones spanning almost the entire P gene. The mRNA, excluding the
poly(A) tail, is 2014 nucleotides long and is bicistronic. The
first open reading frame (ORF) codes for the phosphoprotein (P) of
mol wt 68860. Seven nucleotides downstream from the first AUG
codon, in a +1 reading frame, there is an additional ORF which can
code for a polypeptide of mol wt 23266. The latter protein appears
to be similar to the C proteins found in cells infected with
several paramyxoviruses.
