Lee and Costlow (Methods in Enzymology 152, 633) describe the molecular
titration assay for measuring the prevalence of specific transcripts.

The method involves driving labeled RNA probes into RNA-RNA hybrids as
assayed by nuclease protection.  A constant amount of probe is titrated
with increasing quantities of message.  After nuclease digestion,
hybrids are collected on a filter and counted.  A plot of counts vs RNA
concentration allows one to determine accurately the transcript
prevalence.  The authors claim the method to be the most sensitive RNA
quantitation procedure.
