My understanding of why it is lethal in multi-copy is that if insertion of an AA at a stop codon becomes too efficient, many normal proteins become extended at their C terminus disturbing their function. Thus, raising the gene dosage of the suppressor raises the concentration of the suppressor tRNA causing more efficient read through of the cognate stop codon ultimatly making the cell sick. I can send you a plasmid carrying SUP11 on a BAM Fragment in either YCp50 or YIp5. It is originally from Ron Davis' lab. John R. Thompson -------
