Hello, It is clear that there are plenty of barriers to reproducibility of OFAGE runs. We have recently found that microbial colonisation of the cooling circuit can lead to the destruction of chromosomes, and would be interested to hear from anyone who has dealt with this problem by adding anything to the running buffer (without affecting reproducibility...). I should say that we change our buffer at least every three runs, and flush the system with water before recharging. Even minimal (hardly visible) microbial growth results in bands that are fuzzy.
