Does anyone know of a commercially-available system for purification of DNA binding proteins? I seem to remember seeing one in which the operator region of the DNA of interest is cloned into a pUC (or similar) vector. This recombinant DNA is then attached to an affinity support which consists of lac repressor protein. This complex is then incubated with crude protein extract which contains the DNA binding protein. After washing away unbound proteins,your protein can then be eluted. If anyone knows of such a product, or is familiar with other techniques for partially purifying an unknown DNA binding protein, please pass the information along to me. -------
