Relevant to the problem of restriction sites in close proximity is
rstriction of short DNA sequences. An interesting piece was published
in the NEB transcript December 1989 (New England Biolabs). They have checked
oligos of 8,10,12 or 14 bases with all the most commom enzymes. One example:
Enzyme Oligo Length
BamH I c ggatcc g 8 10% cleavage after 2 hrs
cg ggatcc cg 10 >90%
cgc ggatcc gcg 12 >90%
Might be usefull.
