I feel I have to leap to the defence of vacuum blotting devices somewhat after a recent message. We have been using both Hybaid and Pharmacia devices for something like 18 months for a variety of purposes: Southern blot analysis of Drosophila DNAs, minisatellite analysis of cetaceans and other Mammals, and RNA transfer to name but a few, and have really had very few problems. Indeed the few problems we did have at the beginning were due to us not following the manufacturers' instructions! The problem with high molecular weight material may just reflect the quality of the transfer at high molecular weight! John Hancock, Dept of Genetics, University of Cambridge, UK.
