I'm trying to find the conditions under which the insert in a plasmid can be amplified by adding the bacteria bearing the plasmid directly to a PCR reaction using microtitter plates and a BIOS thermal cycler oven. I've been successfull doing PCR with bacteria in an Ericomb machine. However, there is a limited number of sample wells in the block. Has anybody out there determined the conditions for PCR with bacteria in the BIOS oven using microtitter plates? Any tips will be appreciated.
