[bionet.molbio.methds-reagnts] Southerns with seiving agarose

Jim Gerlach <GERLACHJ@QUCDN.QueensU.CA> (09/05/90)

We are doing some southern blots of PCR'd cDNA and have been separating a
650bp DNA fragment using a gel consisting of 2.75% wide-range agarose (Sigma)
and 1.0% standard agarose (Pharmacia). We seem to be getting poor capillary
transfers to nitrocellulose and wonder if this mix may be contibuting. Has
anyone had experience with transfers from this or similar agarose mixes?

Thanx in advance for any suggestions, comments, etc.

Jim
