I am planning to use asymmetric PCR (with chicken chromosomal DNA as a template) + direct sequencing of the amplification products (preferably with Sequenase) to get some sequence of a gene which is difficult to clone. I have some experience of sequencing PCR products, but not of this particular application. I would like to hear about protocols which other people have found out to work well.
