Hello: I recently came across this protocol for minipreps in the "Current Protocols in Molecular Biology" (big red book) for a plasmid miniprep for DNA sequencing. The procedure is basically an alkaline lysis, followed by a LiCl precipitation. The accompanying comments say that this step precipitates the ssDNA and RNA but not the dsDNA. While this did work, does any one know why this occurs? Please reply to one of the addresses below, post to this board. Thank you for your indulgence (!)
