Greetings. We recently bought an electroporator and have been doing some experiments to optimize it for our standard E. coli host strains. At present we are getting about 2 x 10^7 per microgram for HB101 and 10 times that with MC1061. These numbers apply to two different vectors - a 6.1 kB Dictyostelium shuttle vector and the Bluescript vector. These frequencies are well short of the frequencies one is supposed to be able to achieve.
