Well, if you're going to do a Southern, one way to quantitate the plasmid
is to use known quantities of the plasmid on the same blot. After hybrid-
ization, you can use the known plasmid signals as a calibration curve.
Even without a densitometer, this will give a fairly accurate estimate of
how many plasmid copies are present.
