What follows is a map of pUC18 made conforming to the Plasmid Description Language conventions described in the previous posting. This is a very simple map, but almost everyone has access to a map of pUC18 or pUC19. They are in the back of several biotechnology catalogs, including New England Biolabs and Stratagene. Note that I change widths for the polylinker, and lac and amp genes. To keep it simple, there are no restriction sites in this map. %% Executable code follows: 2686 PLASMIDLENGTH 0 inch WIDTH 2450 235 arcSEGMENT % IG 0.1 inch WIDTH 50 percentBLACK 1750 2450 arcSEGMENT % ampR 0.05 inch WIDTH 0 percentBLACK 580 690 arcSEGMENT % lacI' 235 396 arcSEGMENT % lacZalpha 447 460 arcSEGMENT % lacP 0.025 inch WIDTH 396 447 arcSEGMENT % polylinker 0 inch WIDTH 460 580 arcSEGMENT % IG, lacZ - lacI 690 1750 arcSEGMENT % IG, lacI - ampR 18 POINTSIZE /ZapfChancery-MediumItalic SETFONT (pUC18 ala CW) X Y CenterPText showpage -- --------- Craig Werner (Lognames: werner, craigw) "This is no social crisis, just another tricky day for you."
