Does anyone know of an assembler which will run on a C64 with
one Commodore disk drive?

There is an assembler available on tape, of course
also useable with disk. Its memory usage is less than 4k.
You can obtain it on tape from:

Its price is fl. 60,- excl. postage.
I have used this assembler myself and it functions well.

Believe it or not, the LOGO language for the c64 contains
an assembler written in LOGO.  I haven't used it much, but it works!

As far as I know Commodore themselves produce an assembler called
the Commodore Development Assembler Package costing about 30 pounds.
It includes an editor, monitor and disc assembler.
The package has Macro facilities and does everything a good assembler
should do, including throwing pages at convenient spaces on the
printer so you don't have text along the dotted line!
It's got library files, linked files,etc..
So if you ever get a 150k source prog use this!
